Juglone is a widely known chemical compound. While its toxicity is well established, it still possesses a number of unique qualities. It has been found to be useful in the treatment of malaria and trypanosomiasis (sleeping sickness), because it appears that certain species of the protozoa are sensitive to juglone or other agents capable of producing such sensitivity; in addition, since juglone and its analogs can inhibit some enzymes of eukaryotic cells, they may find use as pharmacological agents for the treatment of various maladies associated with these organisms, including cancer and fungal infections caused by those same organisms. It is well known in the art that certain compounds are useful for inhibiting one or more of the functions of tyrosine kinase enzymes which play an important role in cell proliferation, cancer, atherosclerosis, restenosis, fibrosis, angiogenesis, and other pathological conditions associated with hyperproliferative diseases such as inflammation, immunologic disorders, and autoimmune disease states such as rheumatoid arthritis, psoriasis, Kaposi's sarcoma, and asthma. A method of making a drug useful for treatment of diseases caused by protozoa comprising contacting said protozoa with an effective amount of a compound selected from the group consisting of juglone and derivatives thereof, and a pharmaceutically acceptable carrier therefor so as to render the protozoa more susceptible to drugs known to be effective against the protozoa and thereby to effect an increase in the effectiveness of the latter, whereby diseases resulting from the presence of such protozoa can be treated. Incapsulated juglone matrices can be bound to protein carriers in a variety of ways of which even modern biologists have very distant knowledge. The encapsulation is performed either by a layer-by-layer method, where polyelectrolyte layers are deposited one upon the other or by using multilayers as discussed by Ulbrich et al., (Uhlrich, W.; Asinger, S.; Krapf, M.; Liebner, F.; Langbein, U.; Kohler, H.-P.: Multilayered nanoparticles: preparation, characterization and potential use for controlled release applications, J Am Chem Soc 1995, 117, 6841–6851). Although this technology is based on established methods, it has been introduced only recently in the field of drug delivery and only recently a pilot synthesis was presented. In conclusion, these results demonstrate the possibility of juglone to be toxic and non-toxic simultaneously. These two effects are regulated by a different mechanism that is not related to ROS production or cell death induction, but rather on the change of signaling pathways and gene expression associated with cell cycle arrest, which could be beneficial for cancer therapy, if properly used in combination with other anticancer drugs or radiation therapy.

The cytotoxicity assay was performed using the MTT method, as described by Mosmann et al We first demonstrated that 24 hours incubation with 10 μM juglone or 20 μM thymol could reduce cell viability to 50% or more. To determine whether this is mediated by apoptosis induction, we analyzed the effects of JTMC on the expression of Fas antigen, which plays a key role in apoptotic signaling, in human bladder carcinoma cells and mouse fibroblasts by flow cytometry and Western blot analysis, respectively, after treatment with JTMC for various times (0-48 h). The results demonstrated that JTMC induced apoptosis in both cancer cells and mouse fibroblasts by up-regulation of Fas antigen expression within 24 h.
