Cardiac microsomes were prepared from rabbit heart ventricles using
a protocol similar to that of Tada et al. (1988). Ventricles were
trimmed of fat and connective tissue and homogenized in a small
Waring blender in a buffer containing 10 mM imidazole, pH 7.0, 0.3 M
sucrose, 0.5 mM dithiothreitol (DTT), and the following protease inhibitors:
0.5 mM phenylmethylsulfonyl fluoride, 0.5 mg/ml leupeptin, 0.5
mg/ml aprotinin, 0.5 mg/ml pepstatin, and 1 mM benzamidine. Homogenization
was carried out in three bursts for 15 s, followed by 10 strokes
in a Teflon homogenizer. The homogenates were centrifuged for 10 min
at 4000 g, and the supernatant was filtered through cheesecloth and
centrifuged for an additional 10 min at 27,000 g. Microsomal membranes
were then sedimented by centrifugation of the supernatant for
1 h at 120,000 g. The pellet was resuspended in a solution containing
0.6 M KCl, 0.3 M sucrose, 20 mM Tris maleate, pH 7.0, 0.5 mM DTT, and
protease inhibitors, and the microsomes were sedimented for 1 h at
120,000 g, leaving contaminating contractile proteins in the supernatant.
The resulting pellet was resuspended in storage buffer containing
0.15 M KCl, 0.25 M sucrose, 10 mM Tris maleate, pH 7.0, 0.5 mM
DTT, and protease inhibitors. Aliquots were quick-frozen and stored in
liquid nitrogen. Before use, microsomes were thawed on ice and washed
twice in storage buffer lacking DTT and protease inhibitors.


Можно ли в этой методике используемый набор ингибиторов протеаз заменить вот на это http://www.sigmaaldrich.com/catalog/pro ... =en®ion=BY (отдельно тех ингибиторов, которые приведены в методике у меня нет)?
