Биохимики!

Кто определял СОД с использованием кверцетина и нитросинего тетразолия? Поделитесь рабочими методиками. СОД нужно определить в печени и сердце мыши.

Вот методика (разберетесь наверное) и несколько статей:

1. С. Чевари, И. Чаба, Й. Секей. Роль супероксиддисмутазы в окислительных процессах клетки и метод определения ее в биологических материалах. Лабораторное дело.–1985.– №11. с. 678-681

The role of superoxide dismutase (SOD) in the defense mechanism during intracellular one-electrone reduction of oxygen and formation of superoxide radicals is described. The principle of SOD determination in biological material Is based on SOD ability to compete with nitroblue tetrazoleum for superoxide radicals resulting from aerobic interaction between reduced NADH and phenazinemetasulfate (PMS). Trials of the method included study of the effect of various concentrations of the enzyme, NADH and PMS, temperature, heat inactivation of the enzyme, and kinetics of the reaction. Experimental and computation data have shown that the reaction conforms with the Michaelis enzymatic theory.

Superoxide dismutase (SOD) activity and its isoenzyme spectrum have been studied in the blood plasma and red cells of adults aged 17—35 and in the umbilical blood of the newborns. Electrophoretic analysis has shown thai SOD of the plasma and red cells are not identical: the plasma SOD has 1 fraction and the red cell SOD 3 fractions. This fact underlies the hypothesis that the plasma SOD comes from the tissues and its activity depends on the features of oxygen metabolism in the ceils. The level of SOD in tissues of the newborns seems to be as high as in red cells.

The methods for determination of glutathione reductase and superoxide dismutase activities in human red cells were modified to be fit for biochemical autoanalyzers. The suggested methods will contribute to considerable improvement of the assay accuracy and to reduction of the reagent consumption.

The activities of the red cell superoxide dismutase and glutathione reductase markedly augment in chronic diseases of the liver (persistent and active hepatitis or cirrhosis). Increase of the activities of the above enzymes, important components of the antioxidant system, may be regarded as an adaptive reaction directed at the prevention of lipid peroxidation intensification and its injurious effect on the cellular structures.

Superoxide dismutase (EC 1.15.1.1) activity was assayed in preparations obtained from several clonal lines of nerve cell culture, by enzymatic and nonenzymatic assays. The enzyme was found in dialyzed homogenates of washed cells and in partially purified fractions. The enzyme was also found in cells which had been grown in media containing 5-bromodeoxyuridine, where cell differentiation was observed.

Superoxide dismutase from beef brain and rat liver was assayed in an enzymatic system, using xanthine oxidase, and a non-enzymatie system, based on aerobic reduction of nitro-blue tetrazolium in presence of phenazine methosulphate. The non-enzymatic assay is rapid and simple and permits simultaneous analysis of many samples. Similar results are found by the two methods of assay of superoxide
dismutase.
